Probing c-secretase–substrate interactions at the single amino acid residue level

چکیده
Intramembrane proteolysis by c-secretases plays major roles in disease pathology and cellular signalling, yet the dynamics of these enzyme complexes and how they recognize substrates remains poorly understood. New work in The EMBO Journal utilizes photo-affinity cross-linking to map APP interactions to different c-secretase subunits, suggesting a succession of recruitment and engagement steps that lead up to substrate cleavage.

In this paper, the hydrogen bonding (HB) effects on the NMR chemical shifts of selected atoms in serineand serine-nH2O complexes (from one to ten water molecules) have been investigated with quantummechanical calculations of the 15N and 13C tensors. Interaction with water molecules causes importantchanges in geometry and electronic structure of serine. For the compound studied, the most importan...
متن کاملSubstrate recruitment of c-secretase and mechanism of clinical presenilin mutations revealed by photoaffinity mapping
Intramembrane proteases execute fundamental biological processes ranging from crucial signaling events to general membrane proteostasis. Despite the availability of structural information on these proteases, it remains unclear how these enzymes bind and recruit substrates, particularly for the Alzheimer’s diseaseassociated c-secretase. Systematically scanning amyloid precursor protein substrate...

In this study, solid state fermentation (SSF) was carried out to produce single cell protein (SCP) from sugarcane bagasse using Saccharomyces cerevisiae. The SSF experiment were performed in a tray bioreactor. The influence of several parameters including extraction buffer, initial moisture content of substrate, fermentation time, relative humidity in bioreactor, the bioreactor temperature and ...
